Animals were immunized with 60S spliceosomes purified by gel filtration. Spleen cells were fused with FOX-NY myeloma cells. The antibody reacts with a 35000 dalton protein present in nuclear extracts. On Western blots, the antibody detects a doublet at about 35000 daltons. It can be used to deplete nuclear extracts of splicing activity, to detect mammalian SC35 by Western blot, to precipitate purified spliceosomes and to study mRNA splicing in vitro. |
