Derivation |
The DB cell line was established by Walter J. Urba and Dan L. Longo from ascites fluid from a patient with a diffuse large cell lymphoma. |

The cells do not express HLA-DR, HLA-DQ, IgM , IgD or T cell rantigen receptor.
Exposure of DB cells to phorbol esters (PMA or PdBu) that activate protein kinase C results in profound growth inhibition.
They are reported to be negative for the Epstein-Barr virus genome.
The cells were cured of a prior mycoplasma contamination by a 21 day treatment with BM-cycline. |
Complete Growth Medium |
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.

Subculturing |
Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 x 105 viable cells/mL. Maintain cultures at a cell concentration between 5 x 105 and 2 x 106 cells/mL. Do not allow the cell concentration to exceed 2 x 106 cells/mL.
Medium Renewal: 2 to 3 times weekly. |
