Comments |
The cells are deficient in HGPRT (HPRT) and are resistant to 0.06 mM 6-thioguanine.
The cells remain undifferentiated when cultured on feeder layers (embryonic fibroblasts or STO cells).
The cells spontaneously differentiate and form embryonal structures in the absence of a feeder layer.
When injected into blastocysts, the cells can colonize the germline.
After conventional molecular gene modification techniques, they may be used to reconstitute mouse embryos.

Complete Growth Medium |
The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: 2-mercaptoethanol to a final concentration of 0.1 mM ; fetal bovine serum to a final concentration of 10%.

Subculturing |
Remove medium, add fresh 0.25% trypsin, 0.53mM EDTA solution. Allow the trypsin to loosen the cells, add fresh medium, aspirate and dispense into new flasks containing pre-plated mitomycin-C treated feeder cells (ATCC 56-X.2).
Subcultivation Ratio: A subcultivation ratio of 1:5 is recommended
