Animals were immunized with particulate murine zonae pellucidae. Spleen cells were fused with Sp2/0 mouse myeloma cells. During development and maturation, mammalian oocytes are surrounded by the zona pellucida, which in the mouse is comprised of three sulfated glycoproteins, ZP1, ZP2 and ZP3. The zona pellucida mediates sperm binding at fertilization, provides a post-fertilization block to polyspermy, and facilitates passage of pre-implantation embryos down the oviduct. The antibody specifically binds to mouse ZP2 glycoprotein (amino acids 114-129). The antibody cross-reacts with rat zonae but not with non-rodent species. Following passive immunization, this antibody completely blocks both in vivo and in vitro fertilization of subsequently ovulated mouse eggs. |

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml. Maintain cultures at cell concentrations between 5 X 10 exp4 and 5 X 10 exp5 viable cells/ml. Attached cells may be subcultured by scraping. |
