The growth properties of KHOS-312H are similar to HOS (ATCC CRL-1543).
This line is a revertant of ATCC CRL-1544 that no longer has a rescuable Kirsten murine sarcoma virus genome.
Note: An inconsistency in our records was discovered late in 2010 concerning the human osteosarcoma cell line, KHOS-312H. Due to an ATCC catalog transfer error, this cell line has been referred to as both KHOS-312H and KHOS-321H by ATCC and in the literature. The name originally supplied to ATCC by the depositor was KHOS-312H. |
Complete Growth Medium |
The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing |
Remove medium, rinse with fresh 0.25% trypsin-0.53 mM EDTAsolution, remove trypsin and let the culture sit at 37°C for 10 to 12 minutes or until cells detach. Add fresh medium, aspirate and dispense into new flasks. Subculture every 7 days.
Subcultivation Ratio: A subcultivation ratio of 1:4 is recommended
Medium Renewal: 2 to 3 times per week
