Biosafety Level |
1
Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country. |
Applications |
It also reacts with flagellin from some E. coli H23 and H24 strains, which are antigenically closely related to the H7 antigen. The antibody is specific for E. coli H7 antigen. High titer, high affinity and large volume ascites can be produced in mice injected intraperitoneally with these hybridoma cells. The antibody has been used in ELISA, Western Blot, lateral flow and chemiluminescense assays. Bacterial flagellum expression or motility should be promoted prior to use of this monoclonal antibody to avoid false negative reactions due to lack of target antigen. |
Derivation |
Animals were boosted with partially purified H7 flagellin. Spleen cells were fused with P3X63Ag8.653 mouse myeloma cells. High titer, high affinity and large volume ascites can be produced in mice injected intraperitoneally with these hybridoma cells. |

Comments |
Animals were immunized with whole bacterium antigen from E. coli O157:H7 (ATCC 43895). Animals were boosted with partially purified H7 flagellin. Spleen cells were fused with P3X63Ag8.653 mouse myeloma cells. The antibody is specific for E. coli H7 antigen. It also reacts with flagellin from some E. coli H23 and H24 strains, which are antigenically closely related to the H7 antigen. |
Complete Growth Medium |
Dulbecco's modified Eagle's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and 4.5 g/L glucose and supplemented with 0.1 mM nonessential amino acids, 90%; heat-inactivated fetal bovine serum, 10%
|
Subculturing |
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 5 x 10 exp5 viable cells/ml. Maintain cultures at cell concentrations between 5 X 10 exp5 and 2 X 10 exp6 viable cells/ml. Attached cells may be subcultured by scraping. |
