Comments |
The line is free of major oncogenic avian viruses. The cells are pathogenic in chickens but fluids are not. The cells grow well at 41C. |
Complete Growth Medium |
A 1:1 mixture of Leibovitz's L-15 medium and McCoy's 5a medium with 2 mM L-glutamine, 55%;
tryptose phosphate broth, 5%;
chicken serum, 20%;
fetal bovine serum, 20%
The fetal bovine serum concentration may be reduced to 10% after the cells have been in culture for 10 to 14 days.

Subculturing |
Medium Renewal: Every 2 to 3 days Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 X 10 exp5 viable cells/ml. Maintain cultures at cell concentrations between 5 X 10 exp5 and 2 X 10 exp6 viable cells/ml. |
