Animals were immunized with purified HLA A2. Spleen cells were fused with NS-1 myeloma cells. Mutation at position 107 of the alpha 2 domain of the HLA-A2 molecule results in complete loss of binding of PA2.1. The antibody reactive correlates with the classical HLA A2 specificity. PA2.1 and BB7.2 (ATCC HB-82) recognize overlapping epitopes on the alpha 2 domain of HLA-A2. Tested and found negative for ectromelia virus (mousepox). |
