Cell staining demonstrated intracellular expression of the product of the v-raf gene in these cell lines. Analysis demonstrated the presence of peritoneal macrophage associated cell surface antigens, interleukin-6 (IL-6) secretion induced by lipopolysaccharide (LPS), and biological response modifier-induced cytotoxic activity against tumor cells. The cells are phagocytic, non-specific esterase positive and are Fc receptor positive. The cells produce nitric oxide (NO) when stimulated with a mixture of rMuIFN-gamma and LPS. |

PMJ2-PC (ATCC CRL-2457) and AMJ2-R (ATCC CRL-2458) are cloned, continuous, peritoneal macrophage cell lines generated from C57BL6J mice by in vivo infection with the J2 retrovirus carrying the v-raf and v-myc oncogenes. Cell staining demonstrated intracellular expression of the product of the v-raf gene in these cell lines. The in vivo immortalized cells had many of the morphological and functional characteristics of peritoneal macrophages. Analysis demonstrated the presence of peritoneal macrophage associated cell surface antigens, interleukin-6 (IL-6) secretion induced by lipopolysaccharide (LPS), and biological response modifier-induced cytotoxic activity against tumor cells. The cells are phagocytic, non-specific esterase positive and are Fc receptor positive. One of the clones, PMJ2-PC, constitutively expressed major histocompatibility complex (MHC) class II antigens, and in the other clone, PMJ2-R, MHC class II antigens expression was induced by recombinant murine interferon-gamma. The cells produce nitric oxide (NO) when stimulated with a mixture of rMuIFN-gamma and LPS. |
