Animals were immunized with recombinant human thyroid stimulating hormone (TSH) receptor expressed in. E. coli. Spleen cells were fused with Sp2/0 mouse myeloma cells. The antibody recognizes the N-terminal extracellular domain (amino acids domain 357-389), beta subunit of the human thyroid stimulating hormone (TSH) receptor. It recognizes an extracellular epitope of the beta subunit anchored into the cellular membrane. The antibody does not interfere with hormone binding and has no effect on the receptor mediated adenylate cyclase activity. The antibody cross-reacts with rodent TSH receptor. The TSHR-R5T-34 (ATCC CRL-2683) antibody recognizes a N-terminal extracellular epitope of the beta subunit while the TSHR-T3-365 (ATCC CRL-2684) antibody recognizes a C-terminal intracellular subunit of the beta subunit. The antibodies can be used in Western Blot, ELISA, immunocytochemistry, and immunohistochemistry assays of the human TSH receptor. |
