Applications |
The UC10-4F10-11 hybridoma cell line was produced by fusing Sp2/0-Ag14 myeloma cells with spleen cells from an Armenian hamster that had been immunized with the extracellular portion of of murine CTLA-4 fused to a murine IgG2a. The antibody can be used to detect and quantitate CTLA-4 expression by flow cytometric analysis, and can be used to immunoprecipitate the antigen from CTLA-4 positive cells. Under conditions of suboptimal T cell receptor (TCR) stimulation, whole antibody or Fab fragments from 4F10 can augment T cell proliferation in allogeneic mixed lymphocyte reactions. However, when optimal TCR stimulation is present, 4F10 can suppress T cell proliferation. |

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml. Maintain cell density between 1 X 10 exp5 and 1 X 10 exp6 viable cells/ml. |
