Contains unique BamHI, BglII, XbaI, NruI, and SalI sites, with XbaI and BglII inactivating TRP1. Restriction digests of the clone give the following sizes (kb): BamHI--6.3; BglII--6.3; SalI--6.3; EcoRI--4.6, 1.7. Demonstrates mitotic stability increased over YRp7'; 60-90% of transformed S. cerevisiae J17 maintain the Trp+ phenotype after 20 generations. Constructed by inserting a 627 bp BamHI/Sau3AI fragment containing CEN3 into the BamHI site of YRp7'. |
