Restriction digests of the clone give the following sizes (kb): SacI--4.0, 2.7; EcoRI--6.7; BamHI--6.7; PstI--3.6, 1.9, 0.8, 0.3; HindIII--6.7. Probes I (ATCC 59782/59783), II (ATCC 59784/59785), III (ATCC 59786/59787) and IV (ATCC 59788/59789) constitute non-overlapping fragments of the human von Willebrand factor cDNA sequence that together span essentially the entire 9 kb cDNA. The 2.75 kb FspI/SacI portion of the insert constitutes 'probe III' and corresponds to amino acids 544-1461 of von Willebrand factor. Detects the gene on chromosome 12 and a pseudogene on chromosome 22. Insert is the 5' 4 kb SacI(vector)/SacI fragment of lambda HvWF3. |
