Growth Conditions

Protocol: ATCCNO: 30499 SPEC: This strain is distributed as a frozen stabilate. See general instructions for thawing and storage of frozen material before proceeding. Upon arrival remove the frozen ampoule from the dry ice and transfer directly to a 35C water bath. Inject the entire contents of the ampule intraperitoneally into a golden hamster at least 8 weeks old. Serial passage in golden hamster every 30-45 days. The spleen is homogenized in a balanced salt solution (salt solution for ATCC medium 811 is suitable), approximately 5.0 ml solution/spleen. Inoculate 0.3-0.5 ml of spleen homogenate/hamster. Dosage should be a minimum of 2 X 10(6) amastigotes/hamster. The course of infection will vary with the size of the inoculum.
