Restriction digests of the clone give the following sizes (kb): BamHI--5.1, 3.9, 2.8, 1.6; EcoRI--6.5, 2.7 (doublet), 1.6; HindIII--10.9, 1.8, 0.7; PstI--4.4, 2.2 (doublet), 1.3, 1.2 (doublet), 1.0 + smaller; XbaI--6.8, 4.3, 2.1. The insert contains the following restriction sites (nt position according to published sequence): BamHI--2067, 3633, 7529; EcoRI--2184, 3828, 6430; HindIII--1917, 2569, 2588. The encoded protein binds to the HIV-1 enhancer, the MHC H2-Kb promoter, and the immunoglobulin kappa enhancer. Detects genomic HindIII fragments of the following sizes (approximate kb): human--17, 4.9, 3.0; mouse--10. Extends from the NotI site at nt 152 (blunted) to a SmaI site (blunted) beyond nt 9015. Contains the complete coding sequence and a poly(A) tail of approximately 100 nt. The MspI polymorphism was detected using the 1.6 kb EcoRI fragment. |
