This is an E. coli containing 3 plasmids - pRRS-SacIR+, pLG-SacIM+ and pLG339- that produces the restriction endonuclease and modification methylase SacI. This should be grown on LB media containing ampicillin (50 ug/ml), kanamycin (50 ug/ml) and tetracycline (15 ug/ml) to maintain all plasmids. This culture must be grown at temperatures no greater than 30C to maintain stability. |
