Restriction digests of the clone give the following sizes (kb): BamHI--6.7, 0.5; EcoRI--6.4, 0.8; EcoRV--7.2; SalI--7.2. RP6 is a suicide replicon in hosts not producing the pir protein. RpsL (streptomycin sensitive) is dominant to streptomycin resistance, so loss of the plasmid can identified by streptomycin resistance and ampicillin sensitivity. Suicide, gene replacement vector for Yersiniae and other gram-negative hosts that permits positive selection for loss of the plasmid after allelic exchange. The vector also contains cos sites and oriT, and can be used for cosmid cloning or conjugative transfer. The order of the major features in the plasmid is: EcoRV - R6K ori - oriT - SalI - cos - rpsL - ampR. |
