Restriction digests of the clone give the following sizes (kb): EcoRI--4.0, 2.7; HindIII--6.6; PstI--6.6; BamHI--6.6; PvuII--4.4, 3.2. The insert contains HaeIII, SacI (2), MboII, AccI, and HphI sites. The insert includes the complete exon for the constant region plus 5' and 3' flanking sequences. The polymorphic SacI site is located 3' to the SacI site in the coding sequence of the kappa constant region. Enzyme(s) not detecting polymorphism: ApaI, AvaII, BamHI, BclI, BglII, BstEII, EcoRI, EcoRV, HincII, HindIII, HinfI, KpnI, MboI, MspI, PstI, PvuII, TaqI, XbaI (tested on 7 unrelated Caucasians). |
