Restriction digests of the clone give the following sizes (kb): BamHI/EcoRI--9.3, 8.2, 1.8; NotI--19.3; SstI--19.3. A Ura- yeast strain containing the desired YAC can be transformed with the 8.25 kb EcoRI-BamHI fragment of the vector, followed by selection on uracil-deficient media to form deletion variants of original clone. The NotI and SacI sites are unique and may be used to rescue the right end of the YAC. G418 resistance can also be used to select for transformed mammalian cells. pSE1 (ATCC 87312) and pSE2 (ATCC 87313) differ only in the orientation of the Alu repeat sequence.
