Restriction digests of the clone give the following sizes (kb): EcoRV - 9.4; NdeI - 9.4; SalI - 9.4. The parental plasmid pSR-REP which encodes pol, derived from the Schmidt-Ruppin subgroup A strain of RSV, was described in Cell 34: 597-607, 1983. Rous sarcoma virus (RSV) derived vector (5' LTR, gag, pol and 5' env), for ligation with avian retroviral expression vectors (ATCC 87699 - 87701) to create proviral DNA capable of producing replication competent recombinant virus. The vector can be linearized with SalI for ligation to one of the RSV-derived expression vectors. The pol sequence is derived from the Bryan strain of RSV and is more efficient in virus production after transfection. |
