Restriction digests of the clone give the following sizes (kb): BamHI/EcoRI--4.0, 0.82; EcoRI--4.9; PstI--4.9; RNA transcripts can be generated from the vector T7 promoter following linearization with BamHI. Transcripts undergo autolytic processing. Contains the circularly permuted dimeric tobacco ringspot virus satellite RNA sequence of the pBB1 derivative plasmid pSP651. RNA transcripts contain 723 nt of STobRV RNA sequence (117 nt of promoter-proximal, 247 nt of promoter-distal, and 359 nt of monomeric STobRV RNA) preceded by 21 nt derived from the vector. |
