The plasmid contains the following restriction sites (approximate kb from the pBR322-derived EcoRI site): XhoI--2.5; EcoRI--8.1, 9.1, 11.4, 11.9, 12.6, 15.1, 16; BamHI--14.6, 19.7. Fermentation of lactose in whey GenBank M18108 contains the sequence for the promoter region and part of the coding sequence for LAC4. The vector is a derivative of pBR322 carrying the kanamycin (G418) resistance gene of Tn903 and ARS1B from Kluyveromyces lactis. This plasmid can replicate in E. coli, S. cerevisiae, and K. lactis. The order of the major features in this plasmid is: ampR - kanR - LAC12 - LAC4 > - ARS1B. |
