Restriction digests of the clone give the following sizes (kb): BamHI--10.2; BglII--10.2; PstI--10.2. Cloning into the PstI site inactivates the ampicillin marker used for selection in Escherichia coli. Cloning into the BglII, SphI, or SstI sites inactivates the tyrosinase gene. The pAkij1 ori may be useful in other taxa such as Bacillus, Staphylococcus and other Actinomycetes, including Streptosporangium, Actinoplanes, Nocardia, and Micromonospora. |
