Restriction digests of the clone give the following sizes (kb): PstI--3.0, 1.8, 1.4, 1.1, 0.8; xbaI--uncut; BamHI--3.0, 1.9, 1.7, 1.4, 0.32; HindIII--8.0; EcoRI--8.0. Encodes nt 1-2580 of the published sequence, including the entire coding region and 3' untranslated region and a portion of the 5' untranslated region. When pLDLR-2 is transfected into COS-M6 cells, protein is expressed that binds low-density lipoprotein and that is recognized by monoclonal antibody IgG-C7, specific to the human receptor. The vector contains a portion of pBR322 and SV40 sequences including SV40 ori, 16S and 19S donor and acceptor splicing sites, and polyadenylation signals. Constructed by joining the cDNA insert of p203 (nt 1-323) and p101 (nt 324-5144) via overlapping HgiAI sites. |
