Restriction digests of the clone give the following sizes (kb): BamHI--20.0, 9.0, 5.0, 4.3, 1.8, 1.7, 1.7, 0.7; BglII--25.0, 7.0, 3.2, 3.0, 2.4, 1.5, 0.68; PvuI--20.0, 12.0, 8.0; HindIII--21.0, 18.0, 4.3, 0.8; SmaI--20.0, 19.0, 5.6. The insert contains the following sites separated by (kb)(approx): SalI, MboII- 1.5- BamHI- 1.7- BamHI- 1.7- BamHI- 4.3- BamHI- 1.8- BamHI- 0.65- BamHI- 5.0- SmaI- 0.6- BamHI- 0.7- MboII, SalI. Codominant inheritance was observed in 3 informative families (39 children). The 1.6 kb BamHI fragment contains exon 7 and 3' sequences. Exon 7, which has been sequenced, encodes the last 64 amino acids and includes 1.2 kb of 3' non-coding sequence. The BamHI 1.6 doublet detects the TaqI RFLP associated with DRD2. Use standard hybridization conditions with the addition of human placental DNA to block repeat sequences. Washing conditions are standard with a final wash at 0.1 x SSC, 0.1% SDS, 65C. Enzyme(s) not detecting polymorphism: 31 enzymes. |
