Restriction digests of the clone give the following sizes (kb): EcoRI--2.7, 1.2, 0.9, 0.9; HindIII--4.0, 1.7; SspI--2.3, 2.3, 0.7, 0.3; AvaI--4.3, 1.4; BglI--4.5, 1.2. The insert contains the following restriction sites (approximate kb from the 5' end): HindIII--0.75; EcoRI--1.2; SspI--1.79, 2.04. Because of the internal EcoRI site, the insert cannot be excised intact. For Northern blots, the 1.2 or 0.9 kb EcoRI fragment is recommended. This insert contains two identical copies of 3' 0.9 kb EcoRI fragments. Enzyme(s) not detecting polymorphism: AvaII, BamHI, BanII, BclI, BglII, EcoRI, HindIII, MspI, NruI, PstI, SstI, StyII, XbaI, and XhoI (16 unrelated individuals). |
