EcoRI digest contains fragments of about 20, 11, 6.8, 3.2, 2.6, 1.23, 1.15 and 1.07 kb. Restriction digests of the clone give the following sizes (kb): BamHI--29, 6.4, 5.6, 4.0, 1.6, 0.54; EcoRI--20, 11, 6.8, 3.2, 2.7, 1.3, 1.25, 1.1; HindIII--20, 5.8, 5.8, 4.25, 3.25, 2.7, 2.3, 1.9, 1.15; SalI--uncut; XbaI--20, 15, 12. This genomic clone contains the entire coding region of proenkephalin. To block repeated sequences in the probe, prehybridize with sonicated total human placental DNA instead of salmon sperm DNA. Final stringency wash with 0.1X SSC/0.1% SDS at 65-68C. Enzyme(s) not detecting polymorphism: 29 enzymes including TaqI, MspI, RsaI, BamHI, PvuII, PstI. |
