Restriction digests of the clone give the following sizes (kb): EcoRI--25.0, 13.0, 2.1, 0.7; HindIII--22.0, 9.0, 5.6, 3.8, 2.45; BamHI--18.0, 9.6, 6.0, 5.6, 2.5, 1.4; PstI--12.0, 8.6, 5.6, 4.4, 2.5, 2.3, 1.8, 1.6, 1.2, 0.9, 0.75, 0.5; AvaI--13.0, 9.0, 4.2 (doublet), 3.7 (doublet), 1.8, 1.0. The insert contains the following restriction sites (in approximate kb from the 5' end): BamHI--0.6; BalI--1150; HincII--1375; EcoRI--2150, HindIII--2400. The 3' 0.72 kb EcoRI fragment localizes specifically to 3q26-qter by in situ hybridization. The full length cDNA detects CHE1, CHEL2 and CHEL3 on chromosome 16, but with lower efficiency than chromosome 3. The full-length insert has been used to detect a single 2.4-2.6 kb transcript in human brain and liver. It detects an additional 4.0 kb transcript in human glioblastomas and neuroblastomas (about 10% intensity). |
