Restriction digests of the clone give the following sizes (kb): ClaI/HindIII--2.9, 1.0; BamHI--3.9; EcoRI--uncut; PstI--3.9; BglI--2.1, 1.8. Absence of a transcription terminator 3' to the insert may cause instability of the plasmid after induction, due to interference with plasmid replication. When induced at 42C, interferon gamma (insoluble) can be produced to a level up to 14.7 percent of the total cellular protein. Consists of pPLc28 with the lambda PL promoter, a manipulated ribosome binding site from the E. coli trp attenuator, an engineered ATGTGTTAC sequence, and a sequence corresponding to nt 156-1170 of the sequence record. |
