Restriction digests of the clone give the following sizes (kb): EcoRI--2.7, 1.5; HindIII--3.4, 0.56; BamHI--2.7, 1.0, 0.5; PstI--4.0; EcoRI/HindIII--2.7, 0.8, 0.56. Crry and Cr-2 are within 10 kb of each other and are transcribed in the same direction. Spleen 5 cDNA differs from liver 10 cDNA by having an additional 129 nucleotides between the signal sequence and the nucleotides encoding the first 60 aa repeat. Spleen 5 also does not extend as far in the 5' direction. Liver 10 cDNA (ATCC 63064) and spleen 5 cDNA (ATCC 63065) are clones of alternative splice products of the Crry gene. The mRNAs are transcribed in all tissues examined in a 10 (liver 10) to 1 (spleen 5) ratio. When the complete insert is used as a probe, the following genomic EcoRI fragments are detected in BALB/c mice: 15 kb (chromosome 1), 7.5 kb, 5.6 kb (chromosome 8), 4.8 kb, 4.5 kb, and 1.7 kb. When the 469 bp BamHI/BamHI fragment (containing the extra 129 nucleotides) is used as a probe, the following genomic EcoRI fragments are detected in BALB/c mice: 7.5 kb, 5.6 kb (chromosome 8), and 4.5 kb. Detects mRNA species of 1.9, 2.0, and 2.1 kb. |
