Restriction digests of the clone give the following sizes (kb): PstI--9.8; PvuII--4.4, 3.6, 1.7; EcoRI--7.1, 2.7; BamHI--9.8, HindIII--5.4, 4.4. Digestion with BglII and HindIII together release 4 DNA fragments of 2.5, 1.8, 0.6, and 0.5 kb. The 0.6 and 0.5 kb fragments are unique in the genome and can be used as a hybridization probe to detect a 5 kb mRNA (3' end). |
