This plasmid was constructed by linkering XbaI/Sau3A fragments from the Y chromosome with EcoRI and inserting into the EcoRI site of pJB8. There are EcoRI sites in the insert so the intact insert cannot be released by an EcoRI digest. Restriction digests of the clone give the following sizes (kb): BamHI--11.0; BamHI/XbaI--10.0, 0.94; HindIII--6.4, 4.7; SalI--12.5; EcoRI--5.4, 4.2, 0.66, 0.52. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The single-copy BamHI/XbaI fragment has been sequenced. The single-copy probe (0.9 kb) of the insert can be excised with a BamHI/XbaI double digest. It must be separated from highly repeated flanking sequence by gel electrophoresis. At low stringency (2X SSC, 42C) TaqI detects 4 Y-specific bands which are independently polymorphic and several other Y-specific constant bands. |
