Restriction digests of the clone give the following sizes (kb): AvaI--3.85, 0.69; ClaI--4.4; EcoRI--4.4; SalI--4.4; HindIII--4.4. Contains Sau3A (225), AccI (300), HinfI (345), DdeI (365), and SacII (540) sites (approximate distances in bp from 5' end). Suggest using the AvaI/Sau3A fragment as there are 50 bp of Alu sequences 3' to Sau3A site. NIH 3T3 cells were transformed with DNA from the melanoma NK14 cell line. DNA from secondary transfectants was used to isolate the clone. |
