·凍干首次活化,干粉要全部用完,不能保留,用0.2-0.5m1的培養(yǎng)液或者無菌水溶解,接種在1-2個(gè)平板上,因凍干菌種處于休眠狀態(tài),請(qǐng)勿接種多個(gè)平板,以免因接種量不足而導(dǎo)致復(fù)蘇不成功; |
保存說明 |
1、長(zhǎng)期保存的儲(chǔ)存菌株以冷凍干燥保藏為主,特殊要求的菌種可用其它方法進(jìn)行保存,保存期可長(zhǎng)達(dá)幾年至幾十年。2、半儲(chǔ)存菌株以含20%甘油的肉湯或液體石臘封存的半固體瓊脂為主,在0℃以下保存,保存期為3-6個(gè)月。3、應(yīng)用菌株以瓊脂斜面為主,在4℃保存,保存期為1-3個(gè)月。4、菌種的傳代不可超過6次。視菌種的特性及使用情況,一般儲(chǔ)存菌株每5年傳記代一次,每1-2年從儲(chǔ)存菌株轉(zhuǎn)接至半儲(chǔ)存菌株,半儲(chǔ)存菌株每3-6個(gè)月傳代一次,每1-3個(gè)月從半儲(chǔ)存菌株轉(zhuǎn)接接至應(yīng)用菌株。 |
